Abstract
Previous studies isolated a local alkaliphilic Bacillus species, designated Bacillus sp. G1. The GSS (Genome Sequencing Survey) done then discovered that the alkaliphile also contained genes encoding for antiporter proteins and various enzymes such as proteases and glucanases, among others. Currently an effort is in progress to sequence the whole genome of the organism, so as to fully exploit the vast potential of the alkaliphilic genes and alkalitolerant mechanisms contained within it. For this purpose, the bulk sequence data was separately obtained via 454’s pyrosequencing and organized into many separate contigs and scaffolds. A fosmid library was then constructed with the goal of assembling the detached data into a single complete genome sequence. Approximately 8000 fosmid library clones with insert sizes ranging from 20-50kb were produced, and confirmation of inserts were done via representative fosmid extraction and subsequently restriction digestion.
