Abstract

Nowadays, the demand of new natural and safe pharmaceutical production of immunomodulatory and anticancer properties is very high. For many years it have been reported that Pleurotus ostreatus is characterized by pleuran production, a b-1(1,3/1,6)-D-glucan biopolymer. This polysaccharide has binding affinity to carbohydrate receptors on immune cells and thus have good immunomodulatory activity. When cells cultivated in submerged culture system, they are able to production the pleuran as exopolysaccharide (EPS) in the fermentation medium. In this work, a complete industrial platform was developed to increase the pleuran production using submerged cultivation system in semi-industrial scale. At first, different media formulations were tested for polysaccharide production. The medium of highest EPS production was optimized using statistical approach in shake flask level. After this step, cultivations were conducted in 16-L stirred tank bioreactor for bioprocess industrialization and optimization of different bioprocessing engineering parameters to develop complete industrial platform for this polysaccharide in Semi-industrial scale.
