Abstract
A plasmid-mediated transformation method was developed utilizing the axillary meristem of cotyledon explants of Impatiens balsamina var Dwarf Bush. Low transient transformation frequencies were improved by a series of physical parameters such as helium pressure, target distance, target radius, explants orientation and pDNA concentration as biological parameter. In order to maximize axillary meristem of cotyledon sections saturated with microcarriers, a novel overlapping orientation of explants was utilized. In this way, a transient transformation frequency of 50% was obtained and 2.5% of stable gene expression was achieved after 5 weeks on selective media.
